TWEEN, also known as Polyoxyethylene Sorbitan Fatty Acid Esters, is a group of non – ionic surfactants that have found widespread applications across various industries due to their unique properties.

I. Basic Information

TWEEN typically has a faint, characteristic odor and a warm, slightly bitter taste. Physically, it usually appears as a viscous liquid ranging in color from pale yellow to amber, though this can vary depending on the specific type within the TWEEN series. It is a product formed by a series of chemical reactions. First, sorbitol undergoes dehydration to form anhydro sorbitol. Then, anhydro sorbitol reacts with fatty acids such as oleic acid or stearic acid to generate hexose esters. Finally, under the action of a catalyst, it reacts with ethylene oxide to produce TWEEN.

II. Chemical Properties

Hydrophilicity and Lipophilicity Balance

TWEEN has a unique structure. The polyoxyethylene groups in its molecular structure endow it with strong hydrophilicity, while the fatty acid ester part provides lipophilicity. This balanced amphiphilic nature allows it to effectively reduce the surface tension between water and oil phases, making it an excellent emulsifying agent.

Stability

In general, TWEEN is stable in the presence of electrolytes, weak acids, and weak bases. However, when exposed to strong acids or bases, it will gradually undergo saponification reactions. Additionally, the fatty acid chains in its structure are prone to oxidation over time. TWEEN also has hygroscopic properties, so proper drying may be necessary during storage. Prolonged storage can lead to the formation of peroxides.

III. Applications

Biological Experiments

Emulsifying Proteins: In biological experiments, TWEEN, along with non – ionic detergents like Triton X – 100. can emulsify proteins without disrupting their structures. This is in contrast to ionic detergents such as SDS (Sodium Dodecyl Sulfate), which can break down protein structures. By using TWEEN, the native interactions between proteins can be better preserved.

Blocking Agent: TWEEN – 20. in particular, is widely used as a blocking agent. When performing assays like Western blotting, it helps to block unbound sites on the membrane. This reduces non – specific binding of antibodies, thus enhancing the specificity of the detection. Other common blocking agents include BSA (Bovine Serum Albumin), skimmed milk powder, casein, and gelatin, often used in combination with a dilute solution of TWEEN – 20 (usually 0.05 – 0.1%).

Food Industry

Emulsifier: TWEEN serves as an efficient oil – in – water (O/W) emulsifier in the food industry. It enables the uniform dispersion of various substances, such as fats and oils, in aqueous food systems. For example, in the production of ice cream, TWEEN helps to keep the fat droplets evenly distributed, preventing them from coalescing and ensuring a smooth texture. It is also used in salad dressings to prevent separation of the oil and water phases.

Cosmetics Industry

Emulsification and Solubilization: TWEEN is commonly used in cosmetics to emulsify oils and waxes, allowing them to be evenly dispersed in water – based formulations. This is crucial for products like lotions, creams, and shampoos. It also helps in solubilizing essential oils, making it possible to incorporate them into water – based products and fully utilize their beneficial properties. For instance, in a facial toner with added essential oils, TWEEN ensures that the oils are well – dispersed and can be easily applied to the skin.

Pharmaceutical Industry

Solubilizer: TWEEN can be used as a solubilizer for certain drugs. Some drugs that are poorly soluble in water can be made more soluble by using TWEEN. However, it should be noted that TWEEN has some degree of hemolytic effect, with TWEEN – 80 showing the weakest hemolytic activity among the TWEEN series. Additionally, the aqueous solution of TWEEN may become turbid when heated but will clarify upon cooling, without affecting its quality. However, in solution, it can interfere with the action of some bacteriostatic agents.

IV. TWEEN Series Products

TWEEN is actually a series of products, with different types numbered as TWEEN – 20. TWEEN – 40. TWEEN – 60. TWEEN – 80. etc. Each type has its own characteristics.

TWEEN – 20

Composition and Structure: It is a lauric acid ester, consisting of polyoxyethylene sorbitan monolaurate and a portion of polyoxyethylene bis – sorbitan monolaurate. It has a yellow oily appearance.

Properties and Applications: TWEEN – 20 is relatively mild in nature. In addition to its applications in biological experiments as mentioned above, it is also commonly used in the formulation of personal care products, such as gentle facial cleansers, due to its mild cleansing and emulsifying properties.

TWEEN – 80

Composition and Structure: TWEEN – 80 is an oleate, with an amber liquid appearance.

Properties and Applications: It has stronger emulsifying capabilities compared to some other TWEEN types. In the pharmaceutical industry, it is often used as an emulsifier for injectable drugs. In the food industry, it can be used in products where a more stable emulsion is required, such as in certain types of beverage emulsions.
